The year Key Largo premiered, the Florida Keys were struck by two major hurricanes just a month apart.

Edward G. Robinson and Claire Trevor repeated their roles in a 60-minute radio version for Lux Radio Theatre.

Although Huston had bullied a good performance out of Harry Lewis, cast as one of Robinson's goons, the actor could never equal that performance on his own and eventually left acting to open a hamburger stand on Sunset Boulevard. Eventually it became the 27-restaurant Hamburger Hamlet chain, which he sold in 1987 for $33 million.

In 1981, singer-songwriter Bertie Higgins featured the song "Key Largo," inspired by the film, on his album Just Another Day in Paradise. Lyrics included "We had it all/Just like Bogie and Bacall/Starring in our own late late show/Sailing away to Key Largo." The song reached number one on the adult/contemporary charts while the album was Higgins's only one to crack the top 40.

In the "Vapors" episode of seaQuest DSV the characters played by Ted Raimi and Kathy Evison have a date at an interactive theme park re-creating the world of Key Largo.

Posters of the film are featured in two 2002 episodes of The Sopranos, "Mergers and Acquisitions" and "Whitecaps."
